ABSTRACT
Objective To investigate the effects of omeprazole on the expression and the activity of CYP2C19 in HepG2 cells. Methods MTT assay was performed to screen the concentration range of omeprazole which did not inhibit HepG2 cell proliferation. Real-time quantitative polymerase chain reaction(Q-PCR)were carried out to determine the effects of omeprazole(0,0.1,1 and 10 mg/L)on the expression of CYP2C19 mRNA,Western blot was carried out to determine the effect of(0,0.1,1 and 10 mg/L)on the ex?pression of CYP2C19 protein. After incubation of different concentrations of omeprazole(0,0.1,1,10 and 100 mg/L)with microsomes prepared from insect cells expressing human P450 isozyme,the fluorescence of the samples was detected in a plate reader to analyze if omeprazole inhibited the CYP2C19 activity. Results Omeprazole could down-regulate CYP2C19 mRNA and protein expressions of HepG2 cells in a dose-dependent manner. When the concentration of omeprazole reached 10 or 100 mg/L,the fluorescence intensities obviously decreased,indicating inhibited CYP2C19 activity. Conclusion Omeprazole inhibits the expression and the activity of CYP2C19 in HepG2 cell. When omeprazole is used in combination with drugs activated via CYP2C19 metabolism,their interaction should be considered carefully.

ObjectiveTo evaluate the efficacy of stereotactic body radiotherapy combined with coinstan taneous gemcitabine,and gemcitabine alone for advanced pancreatic cancer.Methods56 advanced pancreatic cancer patients were assigned into observation group,which accepted stereotactic body radiotherapy combined with coinstantaneous gemcitabine 500 mg/m2,d1,d8.Other 50 patients were assigned into the control group which only accepted gemcitabine 1 000 mg/m2,d1,d8,d15.Stereotactic body radiotherapy was delivered with a total dose of 4 000-4 500 cGy in 10 fractions.ResultsCT examinations were carried out 2 months after treatment.The response rate of the observation group and control group was 82％ and 16％ respectively,and the pain relief rate was 67％ and 17％ respectively.The time to progression of the observation group was 14 months,and was better than that of the control group(7.5 months,x2 =7.31,P =0.032).The median survival time of the observation group and control group was 15.8 months and 13.2 months,and the difference had no statistical significance(x2 =3.28,P =0.082).ConcolusionStereotactic body radiotherapy combined with gemcitabine has a better overall response rate and a pain relief rate.It can prolong the time to progression,but can't improve the overall survival.

Objective: To explore behavior problems and family environment of children with video games dependence Method:34 video games dependent children and 34 normal children were assessed with EPQ for children and Achenbach Children's Behavior Checklist Parents of both groups were asked to fill up Family Environment Scale-Chinese Version Result:The P and L scores of EPQ were higher in the study group than in control Social status, school status, withdraw, anxiety/depression, social problems, delinquent problems extroversive behavior problems and the total score of CBCL were also higher in the study group Children with video games dependence had families with lower intimacy, lower expressioness, lower active-reaction orientation and organization Their families had more conflicts than families of normal children Conclusion: Video games dependent children have more behavior problems and poorer family environment
